Yaohaibio’s Full Ecological Recombinant Expression System of Nanobody

Nanobody (Nb), namely VHH (variable domain of heavy chain of heavy-chain antibody), was first reported by Belgian scientists in the journal Nature in 1993. It is a camelaceae (camel, alpaca, alpaca and their close relative species), a single-domain antibody composed of a variable region (VHH) of a natural heavy chain antibody that lacks a light chain. The antibody contains only one heavy chain variable region (VHH) and two conventional CH2 and CH3 regions. The VHH crystal is a cylinder with a diameter of 2.5 nm and a length of 4 nm. The molecular weight is only 15 kDa, so it is also called Nanobody (Nb).

Full ecological recombinant expression system

At present, Yaohabio has established a fully ecological recombinant expression system of nanobody. The existing expression systems include: E.coli prokaryotic expression system, yeast expression system (Pichia pastoris), mammalian cell expression system. We are proficient in using a variety of host bacteria to provide customers with high-quality nanobodies according to the actual requirements of customers.

Engineering bacteria construction of nanobody expression (Pichia yeast, E.coli, or mammalian cells can be selected)

1. E.coli expression system (cytoplasmic or periplasmic spatial expression)

2. Pichia yeast expression system (the methanol-induced expression system PAOX1 or the component expression system PGAP) can be selected

3. Mammalian expression system (expression of nanobody through transient or stable transformant)

1. 1-2 weeks for Escherichia coli (excluding gene synthesis cycles)

2. 2-3 weeks for Pichia pastoris

3. Mammalian: 1-2 weeks for transient and about 2 months for stable transfection

For purified nanobodies: purified antibody detection reports by SD S-PAGE, SE C-HPLC (optional), RP-HP LC (optional) and CE-SDS (optional) purity analysis methods

Low expression purification (label)

Express the constructed nanobody of the engineered strain, and purify 1L of expression volume of protein (label recommended).

2-3 weeks

Large-scale expression purification

Ferment nanobody samples in large scale, and express and purify the fermented nanobody at the same time.
